DIGITAL HOLOGRAPHY APPLIED TO MICROSCOPY


 

A method and apparatus for the numerical reconstruction of digital holograms which allows simultaneous amplitude and quantitative phase contrast imaging. The reconstruction method computes the propagation of the complex optical wavefront emitted by the object and may serve to determine the refractive index and/or shape of the object with an accuracy in the nanometer range along the optical axis. The method requires the adjustment of several reconstruction parameters which is performed automatically. The method also includes a digital method for the correction of the optical aberrations. It finds diverse and important applications in microscopy: surface studies, metrology and biological applications for the determination of cell volumes and refractive index
